Gearing up for LC-MS/MS method development

ABSTRACT: Liquid chromatography tandem mass spectrometry (LC-MS/MS) finds applications in wide range of industries including but not limited to pharmaceuticals/ biopharmaceuticals, food (including dietary supplements, cannabis, and hemp), forensics, clinical (including toxicology), chemical, and environmental. LC-MS/MS is considered the ‘gold standard’ for qualitative and quantitative analysis owing to the specificity, sensitivity, and wide linear dynamic range compared to other techniques. A typical analytical method can include one to several hundred analytes to be detected at low levels in complex matrices. Hence, method development is crucial for generation of a robust and reliable validated analytical method that can withstand inter- and intra- day variabilities between instruments and users. In this presentation, we will discuss crucial aspects of LC-MS/MS method development such as importance of appropriate mobile phase selection, Q1 and Q3 scans, tuning and optimization of the LCMS instrument, and MRM method development. Identifying and overcoming matrix-related signal suppression or enhancement will also be discussed.
